Combidex is our investigational molecular imaging agent consisting of iron oxide nanoparticles for use in conjunction with magnetic resonance imaging (MRI) to aid in the differentiation of cancerous from non-cancerous lymph nodes. Combidex received an approvable letter, subject to certain conditions, from the U.S. Food and Drug Administration (FDA) in June 2000. In September 2004, we submitted a complete response to the approvable letter, which was accepted by the FDA and assigned a user fee goal date of March 30, 2005.

Ferumoxytol, the next-generation product in our development pipeline, is currently in Phase III multi-center clinical trials for use as an iron replacement therapeutic in anemic chronic kidney disease patients, whether or not on dialysis. Exploratory Phase II clinical trials of ferumoxytol for use as a contrast agent in magnetic resonance angiography (MRA) are currently ongoing.

Feridex I.V.О®, our liver contrast agent, is approved and marketed in Europe, Japan, the United States and other countries. GastroMARK, our oral contrast agent used for delineating the bowel in MRI, is approved and marketed in Europe, the United States and other countries.

Todays dismounted infantry soldier carries a back-breaking load, usually 100-140 pounds, and still has insufficient ballistic protection, little defense against chemical and biological weapons, and too many pieces of equipment that don’t work well together. The ISNs challenge is to transform today’s cotton/nylon fatigues and bulky equipment belts to a sleek, lightweight battlesuit that provides everything from responsive armor to medical monitoring to communications and more, in one integrated system.

Nanotechnology fits into this vision in two important ways. First, it offers the potential for miniaturization, a key part of reducing weight. Todays hefty radio worn on a harness might be reduced to a button-sized tab on the collar. And a waterproof poncho could be replaced by a permanent nano-thin coating applied to everything the soldier carries. Secondly, because nanotechnology operates at length scales where classical macroscopic physics breaks down, it offers engineers the potential for creating unprecedented new materials properties and devices. Nanotechnology can solve problems that scientists have been struggling with for decades.

Our core technology is based on the characteristic properties of extremely small, coated superparamagnetic iron oxide nanoparticles. Our core competencies include the ability to design such nanoparticles for particular applications, to manufacture the nanoparticles in controlled sizes and to cover the nanoparticles with different coatings depending upon the application for which they will be used. Our technology and expertise enable us to synthesize, sterilize and stabilize these iron oxide nanoparticles in a manner necessary for use in pharmaceutical products such as iron replacement therapeutics and magnetic resonance imaging (MRI) contrast agents.

Our iron oxide nanoparticles are composed of bioavailable iron that is easily absorbed by the body and incorporated into the body's iron stores. As a result, products using our core technology are well suited for use in intravenous (IV) iron replacement therapy. Additionally, the superparamagnetic characteristic of our products results in nanoparticles that become strongly magnetic when placed in a magnetic field, but lose their magnetism once the field is removed. Therefore, use of our nanoparticles results in magnetic resonance images that increase the information available to the reviewing physicians.

Position Statements & Papers
The NIA will coordinate the generation of position statements and papers in areas relevant to the members. Initially, efforts will focus on the following activities:
Conduct of a technology foresight exercise that will examine current products, current developments and future applications; the output from this study will be an evaluation of the diverse jigsaw of products that currently derive distinctive benefit from their nano-structure/formulation together with a view of potential for nanotechnologies over the next 15-20 years
Establishment of an industry-based perspective on the current nomenclature, threats and opportunities associated with risk-based classification systems, and forecasts for the future; some recommendations will be established as ‘industry-supportable’ recommendations.

Securing the Future of Nanotechnology
The NIA works closely with Government and related bodies, in order to assist the development of policies and regulations, that ensure a secure future for nanotechnology.

Our three main areas of research are:

Hepatic Tissue Engineering
We are interested in understanding the structure/function relationship of the liver to improve cell-based therapy for liver disease. In particular, we focus on how the microenvironment around hepatocytes (the functional cell of the liver) affects cell fate and function. We utilize microfabrication tools (methods developed to manufacture microelectronic circuits) to control and study the role of cell-cell interactions, cell-extracellular matrix interactions, soluble stimuli (e.g. O2), and three-dimensional context on hepatocyte function.

Cell-Based BioMEMS
MEMS is a term used to describe integrated microscale devices that combine electrical, mechanical, and even optical components. Since many biological species are on the same length scale as MEMS devices, the synergy of combining biological systems with MEMS (BioMEMS) can provide the basis for novel, highly instructive biological platforms. We are interested in using arrays of living cells as biosensors and as high-throughput platforms to study fundamental aspects of stem cell biology. Our repertoire of tools includes chemical, topological, fluidic, electrical, and optical manipulation of living cells on chip platforms.

Nanobiotechnology
We are involved in a multidisciplinary effort to develop nanomaterials as tools for biological studies and as multifunctional agents for cancer therapies. By bridging the unique electromagnetic properties of nanomaterials with advances in bioconjugate chemistry, photonics, and phage display we aim to develop ‘intelligent' systems for tumor therapy and biomolecular detection. Our interest centers around nanoparticles and nanoporous materials that can be designed to perform complex tasks such as: home to a tumor, sense changes in cells and tissues, enhance imaging, and trigger the release of a therapeutic payload.

With 300 products in use today, what information is available to demonstrate that nanomaterials do not pose unnecessary risks to human and environmental health? What areas are in need of EHS science? How could risks and environmental impacts of nanomaterials throughout the product life-cycle be minimized with engineering practices to improve product safety and to avoid potential future litigation? Our training course is designed to help you navigate the ever-changing world of nanotechnology.

PuraMatrix is the leading commcial synthetic scaffold for 3D cell culture, cancer biology, drug discovery, tissue engineering, bioproduction, and cell therapy research. PuraMatrix is composed of a simple 16 amino acid peptide monomer 1% weight/volume in tissue culture water, which self-assembles in the presence of salt to form ECM-scale nanofibers, that in turn self-assemble into extremely stable, injectable, biocompatible and biodegradable hydrogels. The gels work by coating surfaces or encapsulating cells, allowing for the proper three dimensional (3-D) culture across most cell types, including cell-based assays for neurite outgrowth, angiogenesis, cell migration, cancer invasion, and hepatocyte toxicity. This reproducible integrin-free scaffold can be customized according to the desired microenvironment to closely model the in vivo milieu along with growth factors, cytokines, integrin sequences, and other bioactive molecules.

The Nanoscience Centre is a new building occupying a site at the north east corner of the Cavendish Laboratories. The main activity in the building is making individual devices or structures which are only a few nanometres in size and then measuring how they work. The building was completed in early 2003.

The building is of one storey throughout. Offices for researchers and technical support, together with a small meeting room have been designed to be set into the berm so as to reduce their visibility from Clerk Maxwell Road. To the west of the office area is a laboratory which is split into a clean room facility for the fabrication of the nanometre scale devices and structures, and laboratory space for characterisation of the same. The size of these devices is already much smaller than the dust particles filling the air around us.

Air in the clean room is forced through fine filters in the ceiling of the building and recirculated along with a percentage of fresh air. The filtered air in the clean room prevents contamination of the devices by the atmospheric dust. Within this room various types of patterning and fabrication will be carried out.

Our primary mission is to develop nano-fabrication, advanced lithography and precision engineering technology for building high performance space instrumentation, including x-ray telescopes and high resolution x-ray spectrometers, magnetospheric imagers and solar physics instrumentation.

Our lab is also a leader in the field of nanometer and sub-nanometer dimensional metrology applied to planar lithography patterning and ultrahigh accuracy optics.

Education and training of students is an important part of our mission. Research in the SNL is performed primarily by graduate and undergraduate students from the Departments of Mechanical, Electrical, Aero/Astro Engineering, and Physics with support from a small professional staff.
