Since commencing operations in 2002, Ablynx has generated Nanobody™ leads against a number of human disease targets across a wide range of therapeutic areas. Two of these leads are now in advanced pre-clinical development, and Ablynx’s first therapeutic program is expected to enter human clinical trials in 2006.

Ablynx’s Nanobody™ technology was originally discovered at laboratories of the Free University of Brussels (VUB). Scientists at the VUB discovered that up to half of the antibody repertoire in camelidae consists of antibodies that lack a conventional antibody light-chain, but retain full antigen-binding capacity. The technology and intellectual property created around this discovery was further developed within the Flanders Interuniversity Institute for Biotechnology (VIB). In 2001 VIB created Ablynx in collaboration with founding venture investor, GIMV. Ablynx now operates from its state-of-the-art facilities in Ghent, Belgium.

If the wound is too dry it donates moisture and if it is too wet it absorbs excess moisture to provide a healthy environment for healing wounds.

In addition to this moisture management capability, Microlattice matrix is also an optimal vehicle to deliver active agents to the body over an extended time. A wide range of active agents have successfully been integrated into Microlattice to produce devices that deliver therapeutically significant quantities of agents ranging from elemental oxygen to protein growth factors. The first commercially available products using this approach are the SilvaSorb® silver antimicrobial dressings and gel which incorporate AcryMed’s proprietary stabilized silver chemistry. SilvaSorb is sold in North America by Medline industries and in China by Guongdong Pharmaceuticals Import and Export Corporation. AcryMed is currently seeking additional geographic distribution partners for SilvaSorb.

AcryMed’s scientists have developed and patented two other silver antimicrobial technologies for treating medical devices by direct incorporation or surface impregnation. The company is currently investigating partnership opportunities with medical device companies desiring a state of the art antimicrobial claim.

Our three initial product candidates are designed to address large unmet clinical needs within cardiology, oncology and asthma. Our lead product candidate is a cardiovascular drug in Phase III clinical development for the detection of coronary heart disease, the leading cause of death in the United States.

We created our three initial product candidates using technology that enables us to control the size and porosity of microparticles in a versatile manner, so we can customize the microparticles to address the delivery needs of a variety of drugs. We are focused on creating porous microparticles that are smaller than red blood cells. Combidex is our investigational molecular imaging agent consisting of iron oxide nanoparticles for use in conjunction with magnetic resonance imaging (MRI) to aid in the differentiation of cancerous from non-cancerous lymph nodes. Combidex received an approvable letter, subject to certain conditions, from the U.S. Food and Drug Administration (FDA) in June 2000. In September 2004, we submitted a complete response to the approvable letter, which was accepted by the FDA and assigned a user fee goal date of March 30, 2005.

Ferumoxytol, the next-generation product in our development pipeline, is currently in Phase III multi-center clinical trials for use as an iron replacement therapeutic in anemic chronic kidney disease patients, whether or not on dialysis. Exploratory Phase II clinical trials of ferumoxytol for use as a contrast agent in magnetic resonance angiography (MRA) are currently ongoing.

Single molecule biophysics lies at the confluence of molecular medicine and nanotechnology. The Center uses nanotechnology to study physical processes on which life is based using the simplest model systems — those that exist on the level of a single molecule or several molecules. By doing this, researchers plan to gain a better understanding of gene regulation, molecular signaling and molecular transport in cells that will lead to improved biosensors and other new technologies.

The Center for Single Molecule Biophysics seeks to better understand the physical basis of life by studying the individual molecules that comprise living cells. Researchers at the Center are using scanning probe methods to measure and manipulate individual molecules. Our premise is that single molecules are simple enough to be molded accurately with computer simulations, yet complex enough to reflect the variations that are important to their function.

The Center is also dedicated to advancing the latest techniques for research on the single molecule scale and to translating discoveries into new tools and new applications that are relevant to promoting health and combating disease. With a clearer understanding of the individual components that comprise integrated living systems, researchers are better able to grasp the many complex interactions between gene products (molecules) that cause disease. By learning from nature’s repertoire of experiments on its own ”nanomachines” researchers at the Center are figuring out how to craft components for machinery of undreamt-of complexity and sophistication.

Our research program is inherently interdisciplinary. The tools are in the domain of engineering and chemistry: nanostructures that connect to molecules and instruments that manipulate and measure at the single molecule level. The interactions and modeling come from physics: understanding the quantum phenomena that dominate interactions on this length scale and building simple models of the complex many-particle phenomena. The systems come from biology: molecules involved in gene expression (signaling and the control of processes in living cells). Finally, the building of molecular model systems and the controlled assembly of nanostructures is dependent upon chemistry, biochemistry and materials chemistry).

The recent evolution in nanotechnology has given us new tools with which researchers can imagine measure, manipulate and modify individual molecules, one at a time. Previous probes of biological molecules, such as X-ray crystallography, inferred their properties by averaging billions of molecules. However, single molecule studies now show that, outside the confines of a crystal, proteins fluctuate through a series of very different structures. Therefore, the crystal structures are often not only inappropriate, but they fail to capture the set of states that the protein uses to do its job.

For example, adhesion proteins must adopt one structure when a cell rests and another (non-sticky) structure when the cell moves. But molecules are in a state of constant fluctuation, with, on average, more of them in the ‘sticky’ state when the cell rests, changing to more in the ‘non-sticky’ state when the cells move. The only way to capture the structural changes that drive the process is to study just one molecule as it jumps between the two states. Thus, single molecule methods are often the only way to follow the changes in structure that rival the fundamental processes of life.

The seven-presentation course runs approximately three hours. Each presentation can be viewed separately and ranges from seven to 35 minutes in length. The course introduces fundamental concepts, including changes in the properties of materials at the nanoscale, and the potential of nanotechnology and its application to biomedical sciences. A consortium led by QinetiQ Nanomaterials Limited (QNL), a wholly owned QinetiQ subsidiary, is to develop a range of nanomaterials for use in the fight against a broad spectrum of viruses, it was announced today. The two year programme is being funded by the South East England Development Agency (SEEDA) which has allocated £2m to the project, their largest single investment in science and technology to date.The initial concept was formulated by Dr Guogang Ren at Queen Mary's University of London who discovered that at the nanoscale, certain inorganic materials kill viruses such as SARS and avian flu on contact. Team AVNP, of which Dr Ren is a partner, will develop combinations of nanoparticles and nanocompounds for application to a range of antiviral products such as face masks, air filters and antiviral coatings, making full use of the larger surface area available at the nanoscale. The programme will optimise the effectiveness of nanomaterials and assess their toxicology for different antiviral applications. In addition Team AVNP will restructure the production processes of nanomaterials with the aim of delivering pre-production prototypes.

Key to the programme is the team's unique combination of capabilities in nanomaterials, polymers, toxicology and virology testing that are provided by QNL, Brunel University, Queen Mary's, and Retroscreen Virology Ltd. To develop the applications the Pall Corporation will provide expertise in filtration and Sun Chemical in coatings technology.

Speaking on behalf of Team AVNP, QNL founder and CTO Dr Paul Reip said: "H5N1 Bird Flu is just one of a number of viruses such as SARS that represent a genuine threat across the globe. There is a long term requirement for new and innovative antiviral materials that can be applied to products such as face masks to protect not only frontline staff such as nurses, doctors and paramedics but also the general public.

"Through its most substantial investment in science and technology to date, SEEDA has recognised the strengths of the AVNP team and the potential of nanoparticles to tackle a broad spectrum of viruses.

"The AVNP programme represents a new chapter in the ongoing story of the nanotechnology revolution. Most of the current applications of nanotechnology involve nanopowders, for example using the properties of UV absorption to give UV resistance in sunscreens. This project will exploit additional properties that materials have at the nanoscale to tackle real world crises such as pandemics."

Funding for the programme was provided under a SEEDA programme entitled "Emerging Nanomaterial Technologies in the South East Region." The programme supports new processes and materials in the new field of nanotechnology, and specifically the near to market area of nanomaterials.
